Anti leukemia effect of polygonum cuspidatum extract resveratrol

Leukemia is a common malignant disease of hematopoietic tissue. It was originated in the gene mutation of hematopoietic stem cell. Chemotherapy is the primary mean in treatment of this disease. However, the chemotherapy drugs often kill normal cells in the treatment of leukemia. It causes functional or organic injury to normal tissues and organs of the body, especially for bone marrow and digestive tract. In addition, the tolerability of chemotherapy drugs is also one of the main reasons for failure of chemotherapy. Therefore, there is an urgent need to find a better way for the treatment of leukemia and systemic tumor.
Trans-resveratrol obtained from Shaanxi, China. It is a Chinese medicine from polygonum cuspidatum. It has a higher extraction rate of active ingredient resveratrol. The Resveratrol chemical name of the stilbene phenol, is a non-flavonoid polyphenols, plant resistance to outside stimulus such as ultraviolet light, fungus, virus infection or mechanical damage, a phytoalexin. Although the anti-tumor effect of resveratrol have been confirmed in many cell lines, but there are large differences in the specific application of dose in the different studies.
In vitro cell culture, and observed the effect of different concentrations of resveratrol on the cell. Resveratrol treatments, the cells in each group are different degrees of morphological changes and apoptosis. Resveratrol growth inhibition with strong pro-apoptotic effects. And resveratrol in play at the same time on the cytotoxicity of leukemia cells does not affect the growth of normal cells. In view of the anti-tumor chemotherapy drugs on normal cells tend to cause non-specific killing of resveratrol extracted from Chinese herbal medicine or natural plant not only has good anti-tumor effect, but no obvious cytotoxic effect on normal cells. Therefore, the immune deficiency crowd even more important value.
A growing number of studies have found that a considerable portion of the blood system the occurrence of diseases related to abnormal regulation of hematopoiesis. Leukemia is a malignant clonal disease of hematopoietic stem cells, the path of sustained activation of the abnormal proliferation of leukemic cells to apoptosis blocked and is closely related to differentiation obstacles. The STAT family is a group of DNA binding proteins, and tyrosine phosphorylation of signaling pathways coupled to play a transcriptional regulatory role, mediated by a variety of biological effects, play an important role in the process of tumor development. The study confirmed that activation of STAT3 signaling to the tyrosine kinase signal integration, combined with DNA components, the activation and proliferation-related gene transcription, or increase the expression of antiapoptotic proteins, thus affecting cell survival and proliferation, eventually lead to cancer. Resveratrol can pSTAT3 expression in a dose-dependent weakening of cell. Prompt it is blocked by down-regulating the activity of STAT3 tyrosine phosphorylation, its downstream target gene protein Bcl2 expression, thus it inhibits the growth of leukemia cell proliferation and promotes apoptosis.
Many anticancer pharmaceutical raw materials can induce tumor cell cycle arrest. They suggest cell cycle arrest can play a role in tumor cell apoptosis. Resveratrol plays a inhibiting and proliferation role on leukemia cells through interfering with DNA replication. In different cell lines, the ways that resveratrol plays its role are not the same. At present, for how resveratrol affect the cell cycle, scientists from various countries has not yet reached a consensus. Therefore, it is necessary to further study and reveals the exact molecular mechanisms of resveratrol’s anti-tumor effect.
